چکیده

Sarcoplasmic reticulum (SR) Ca release in intact ventricular myocytes is the major source of Ca which activates cardiac contraction (although Ca influx makes a non-negligible contribution in most species). The fundamental events of SR Ca release are known as Ca sparks. The twitch Ca transient is composed of approximately 10,000 Ca sparks occurring in a given cell, and they are synchronized by the action potential and Ca current. Many factors influence SR Ca release amplitude and kinetics, and the focus here is on understanding how these factors work in the intact cellular environment. The intracellular [Ca] ([Ca]i) and intra-SR [Ca] ([Ca]SR) are two of the most important dynamic modulators of SR Ca release. Indeed, while [Ca]i (and Ca current which initiates systolic SR Ca release) is widely acknowledged to be important, it is increasingly clear that [Ca]SR changes dynamically under physiological conditions and that this has very important regulatory effects on the SR Ca release process. While elevation of [Ca]SR obviously increases the driving force and amount of SR Ca available for release, it also increases the fractional release and can be responsible for spontaneous diastolic SR Ca release. These issues are discussed in both normal physiological and pathophysiological contexts.

Effect of acidosis on Ca2+uptake and release by sarcoplasmic reticulum of intact rat ventricular myocytes.

The effect of acidosis on Ca2+ uptake and release by the sarcoplasmic reticulum (SR) of rat ventricular myocytes has been investigated. Intracellular Ca2+concentration ([Ca2+]i) was monitored using fura 2; the L-type Ca2+ current ( I Ca) was monitored using the perforated patch-clamp technique. Acidosis was produced either by superfusing the cells with an acid solution (intracellular and extrac...

Effects of the Ca2+ sensitizer EMD 57033 on intracellular Ca2+ in rat ventricular myocytes: relevance to arrhythmogenesis during positive inotropy.

We have investigated the effects of the calcium-sensitizing inotropic agent EMD 57033 on Ca(2+) handling in intact and skinned rat ventricular myocytes. Intracellular Ca(2+) was monitored using fura 2. Myocytes were saponin-skinned, allowing study of sarcoplasmic reticulum (SR) function. In intact myocytes EMD 57033 (1-10 micromol/l) produced a concentration-dependent decrease in the amplitude ...

Sarcoplasmic reticulum Ca(2+) release by 4-chloro-m-cresol (4-CmC) in intact and chemically skinned ferret cardiac ventricular fibers.

The purpose of this study was to determine whether 4-chloro-m-cresol (4-CmC) could generate caffeine-like responses in ferret cardiac muscle. The concentration dependence of 4-CmC-mediated release of Ca(2+) from the sarcoplasmic reticulum was studied in intact cardiac trabeculae and saponin-skinned fibers in which the sarcoplasmic reticulum was loaded with Ca(2+). In intact and saponin-skinned ...
